Complement Factor H-related protein 2 (FHR-2) plays an important role in complement regulation. Predominantly synthesized in the liver, but monocytes, macrophages, and dendritic cells also contribute. With a molecular weight of approximately 29 kDa, FHR-2 closely resembles Complement Factor H (CFH), a key regulator of the complement system. Although FHR-2 shares structural similarities with Complement Factor H (CFH), its precise function is still unclear.
Research indicates that FHR-2 may inhibit the alternative pathway (AP) C3 convertase, potentially reducing membrane attack complex (MAC) formation. Conversely, some studies indicate that FHR-2 might instead promote AP activation. Findings support this, as a hybrid mutant protein with FHR-5 domains and full-length FHR-2 lead to overactivation of the complement system.
FHR-2 is linked to various diseases driven by complement dysregulation, such as atypical hemolytic uremic syndrome (aHUS) and age-related macular degeneration (AMD). Its involvement in the alternative pathway makes it a promising therapeutic target for managing complement-driven diseases.
